目的:探讨切断左侧迷走神经对大鼠血清降钙素基因相关肽含量和降钙素基因相关肽(CGRP)基因表达及调控机制的影响。方法:30只SD大鼠切断左侧迷走神经并按术后取样时间随机分为术后1周组、术后1个月组、术后3个月组,每组10只;另设10只SD大鼠作为正常对照组。采用放射免疫分析法测定血清中CGRP浓度,RT-PCR法测定肺内CGRPmRNA表达。结果:迷走神经切断术后1周和1个月血清CGRP含量比正常对照组显著降低(P﹤0.01),随着时间延长,血清中CGRP含量逐渐增加,至3个月时接近正常对照组(P﹥0.05);而肺组织CGRPmRNA表达在术后1周和1个月均明显高于对照组(P﹤0.01),随着时间的延长,其表达量逐渐减少,至术后3个月时接近正常对照组(P﹥0.05)。结论:迷走神经切断后血清内CGRP浓度下降及肺内CGRPmRNA表达量增加,表明迷走神经对CGRP基因的表达和神经活性物质CGRP的分泌具有调节作用。
Objective: To investigate the effect of cutting off the left vagus nerve on the expression of calcitonin gene-related peptide and the expression of CGRP in rats. Methods: Thirty SD rats were sacrificed and the left vagus nerve was excised. The rats were randomly divided into 1 week group, 1 month postoperative group and 3 months postoperative group, 10 rats in each group. Another 10 SD rats Rats as normal control group. Serum CGRP levels were measured by radioimmunoassay, and lung CGRP mRNA expression was determined by RT-PCR. Results: After 1 week and 1 month after vagotomy, serum CGRP level was significantly lower than that of the normal control group (P <0.01). With the prolongation of time, the level of CGRP in serum gradually increased and reached the normal level at 3 months (P > 0.05). The expression of CGRP mRNA in lung tissue at 1 week and 1 month after operation was significantly higher than that in the control group (P <0.01). The expression of CGRP decreased gradually with time, reaching to 3 months after operation Normal control group (P> 0.05). CONCLUSION: The decrease of serum CGRP concentration and the increase of CGRP mRNA expression in the lung after vagotomy may indicate that the vagus nerve regulates the expression of CGRP gene and the secretion of CGRP.
