目的利用体外培养的大鼠乳鼠心肌细胞,探讨葛根素(puerarin,Pue)对PKC通路的激动剂(佛波醇酯,PMA)所致心肌细胞肥大的抑制作用和与蛋白激酶C(PKC),c-fos之间的关系。方法新生大鼠心肌细胞培养,除对照组外,分别给予PMA(均为10-7mol.L-1)、PMA+Pue(其中Pue按浓度分为10-8mol.L-1,10-7mol.L-1,10-6mol.L-1)、PMA+10-5mol.L-1白屈菜季氨碱(chelerythrine,Che),观察Pue对培养心肌细胞体积、总蛋白质含量3、H-亮氨酸的掺入、PKC活性及c-fos的影响。结果 PMA组的上述指标与对照组相比显示心肌肥厚模型制备成功(P<0.01);Pue可显著减少肥大心肌细胞的蛋白质含量、心肌细胞体积3、H-亮氨酸的掺入、PKC活性及c-fos的表达,且与che无明显差异。结论 Pue抑制心肌肥大可能与PKC和c-fos有关。
