[目的]探讨泽兰乙醇提取物的舒张血管效应及其机制.[方法]利用多通道生理记录仪对大鼠离体胸主动脉进行观察,观察泽兰乙醇提取物对血管的作用,并从血管内皮细胞及一氧化氮(NO)/环磷酸鸟苷(cGMP)信号通路探讨其作用机制.[结果]泽兰乙醇提取物对内皮完整的大鼠胸主动脉环具有剂量依赖性舒张血管效应.去除血管内皮,可明显抑制泽兰的舒张血管效应;应用一氧化氮合酶抑制剂和鸟苷酸环化酶抑制剂预处理后,泽兰的舒张血管效应明显被阻断;应用盐酸普萘洛尔或阿托品对泽兰的舒张血管作用无影响.[结论]泽兰乙醇提取物的舒张血管效应存在内皮依赖性,且可能与NO/cGMP信号通路相关.
[Objective] To investigate the vasodilative effect of ethanol extract of Zeeland and its mechanism. [Methods] The isolated rat thoracic aorta was observed by multi-channel physiology recorder. The effect of ethanol extract of Zeeland on blood vessel was observed. The effect of ethanol extract of Zeeland on the thoracic aortic rings of endothelium was dose-dependently vasodilated .The removal of vascular endothelium significantly inhibited the vasodilative effect of Zeeland, and the preconditioning of nitric oxide synthase inhibitor and guanylate cyclase inhibitor significantly blocked the vasodilatation of Zeeland. No significant effect of alexine or atropine on the vasodilatation of Zeeland was observed. [Conclusion] The vasodilator effect of ethanol extract of Zeeman is endothelium-dependent and may be related to NO / cGMP signaling pathway.
