目的观察胰岛素抵抗高血压大鼠血小板L-精氨酸/一氧化氮系统的改变。方法自发性高血压大鼠(SHR)自第5周至第10周喂食果糖,制备胰岛素抵抗高血压大鼠(IR)模型。在此模型上,检测血小板一氧化氮合酶(NOS)活性、一氧化氮(NO)产生量及L-精氨酸(L-Arg)转运特征,同时观察了血浆L-Arg水平、心钠素(ANP)、NO水平及cGMP水平。结果SHR大鼠收缩压(SBP)(P<0.01),血浆ANP(P<0.01)和cGMP(P<0.05)水平均较WKY对照组明显升高,而血糖和血浆胰岛素水平无显著变化(P>0.05)。IR大鼠的SBP(P<0.01),血浆ANP(P<0.05)和cGMP(P<0.05)水平较SHR进一步升高;且血糖与血浆胰岛素水平升高明显(P<0.01),三组间血浆L-Arg水平并无明显差异(P>0.05)。IR大鼠血小板NO产生量、NOS活性及血浆NO水平分别较SHR降低25%(P<0.01)、24%(P<0.01)和29%(P<0.01)。L-Arg最大转运速率SHR是WKY的60%(P<0.01)、IR又为SHR的82%(P<0.01),三组间米氏常数(Km)的变化无统计学意义(P>0?
Objective To observe the changes of platelet L-arginine / nitric oxide system in insulin resistance hypertensive rats. Methods Spontaneous hypertensive rats (SHR) were fed fructose from week 5 to week 10 to prepare insulin resistance hypertensive rats (IR) model. On this model, the activity of nitric oxide synthase (NOS), the production of nitric oxide (NO) and L-arginine (L-Arg) transporter were detected. The levels of plasma L-Arg, (ANP), NO levels and cGMP levels. Results SBP (P <0.01), plasma ANP (P <0.01) and cGMP (P <0.05) levels in SHR rats were significantly higher than those in WKY control group, while the levels of plasma glucose and plasma insulin No significant change in level (P> 0.05). SBP (P <0.01), plasma ANP (P <0.05) and cGMP (P <0.05) levels in IR rats were higher than those in SHR group, and the levels of blood glucose and plasma insulin increased significantly (P < 0.01). There was no significant difference in plasma L-Arg level among the three groups (P> 0.05). Compared with SHR, NO production, NOS activity and plasma NO level in IR rats decreased by 25% (P <0.01), 24% (P <0.01) and 29% (P <0.01), respectively.
