目的:研究SLP-2在宫颈鳞癌组织中的表达及其与临床病理参数之间的关系。方法:收集郑州大学第二附属医院经手术切除的组织42例宫颈鳞癌组织、43例宫颈上皮内瘤变(CIN)患者的宫颈组织和48例正常宫颈组织,采用半定量RT-PCR技术检测SLP-2mRNA的表达情况,以上组织在病理科存档的石蜡切片采用免疫组化SP法检测SLP-2蛋白的表达情况。结果:PT-PCR法检测结果显示,与CIN及正常宫颈组织相比,SLP-2mRNA在宫颈鳞癌组织中的表达水平显著降低,F=3 979.485,P<0.01;免疫组化染色结果显示,SLP-2蛋白在正常宫颈组织中的表达率81.3%,CIN组织中的表达率为51.2%,宫颈鳞癌组织中的表达率(14.3%)明显低于CIN及正常宫颈组织,χ2=40.197,P<0.01;SLP-2的低表达与宫颈鳞癌的组织分化程度相关(P=0.033),而与患者年龄、临床分期、淋巴结转移、肌层浸润深度及肿瘤大小无相关性,P值均>0.05。结论:SLP-2在宫颈鳞癌中低表达,SLP-2的低表达在宫颈鳞癌的发生发展中发挥重要的作用,可能与宫颈鳞癌的组织分化程度密切相关。
Objective: To investigate the expression of SLP-2 in cervical squamous cell carcinoma and its relationship with clinicopathological parameters. Methods: Twenty-four cases of cervical squamous cell carcinoma and 43 cases of cervical intraepithelial neoplasia (CIN) from the Second Affiliated Hospital of Zhengzhou University were collected for cervical tissue and 48 cases of normal cervical tissue. Semi-quantitative RT-PCR was used to detect SLP-2mRNA expression, the above tissues in the pathology archived paraffin sections using immunohistochemical SP method to detect SLP-2 protein expression. Results: The results of PT-PCR showed that the expression of SLP-2 mRNA in cervical squamous cell carcinoma was significantly lower than that in CIN and normal cervical tissues (F = 3 979.485, P <0.01). The results of immunohistochemistry showed that, The expression rate of SLP-2 protein in normal cervical tissue was 81.3%, in CIN tissue was 51.2%, in cervical squamous cell carcinoma tissue (14.3%) was significantly lower than that in CIN and normal cervical tissue (χ2 = 40.197, P <0.01. The low expression of SLP-2 was correlated with the histological grade of cervical squamous cell carcinoma (P = 0.033), but not with age, clinical stage, lymph node metastasis, depth of myometrial invasion and tumor size > 0.05. CONCLUSIONS: SLP-2 is low expressed in cervical squamous cell carcinoma and low expression of SLP-2 plays an important role in the occurrence and development of cervical squamous cell carcinoma, which may be closely related to the degree of tissue differentiation of cervical squamous cell carcinoma.
