目的以乳腺癌细胞株MCF-7为靶细胞,考察葛根素对其细胞形态及增殖的影响。方法以人乳腺癌细胞株MCF-7为靶细胞,以雌酚酮1×10-8mol/L为阳性对照,同时设空白对照组,葛根素干预组分为1×10-8mol/L、1×10-7mol/L、1×10-6mol/L三个浓度组。①调整细胞浓度为4×104个/L,接种于12孔培养板上。48h后,倒置显微镜下观察细胞形态变化。②调整细胞浓度为2×104个/L,接种于96孔培养板,采用四唑盐法测定各孔的吸光度,计算平均增殖率。上述实验每组均设8个复孔。③调整活细胞浓度为0.9×104个/ml,接种于24孔培养板。24h后,每组设12个复孔。每天随机从每组细胞中各选出3个孔,染色后在光镜下计数活细胞数,取平均值。连续计数3天,以时间为横坐标,活细胞数为纵坐标,作图得细胞生长曲线。结果经葛根素1×10-7mol/L、1×10-6mol/L处理后第1天、第2天和第3天,与空白对照组相比,增殖速度均加快。结论葛根素可剂量依赖地促进MCF-7细胞的增殖,提示其具有弱雌激素样作用。
Objective To investigate the effect of puerarin on cell morphology and proliferation of breast cancer cell line MCF-7. Methods The human breast cancer cell line MCF-7 was used as the target cells and the estrone 1 × 10-8 mol / L was used as a positive control. The blank control group was treated with 1 × 10-8 mol / L puerarin × 10-7mol / L, 1 × 10-6mol / L three concentration groups. ① adjust the cell concentration of 4 × 104 个 / L, seeded in 12-well culture plate. After 48h, the morphological changes of the cells were observed under inverted microscope. ② adjust the cell concentration of 2 × 104 / L, seeded in 96-well plates, the use of tetrazolium salt method to determine the absorbance of each well to calculate the average rate of proliferation. The above experiment each set eight complex holes. ③ adjust the live cell concentration of 0.9 × 104 个 / ml, seeded in 24-well culture plate. After 24h, each set of 12 complex wells. Three randomly selected wells from each group of cells were randomly selected each day. After staining, the number of viable cells was counted under a light microscope and averaged. Continuous counting 3 days, with time as abscissa, the number of living cells for the vertical axis, the cell growth curve was plotted. Results Compared with the blank control group, the proliferation rate increased on the 1st day, the 2nd day and the 3rd day after treatment with puerarin 1 × 10-7mol / L and 1 × 10-6mol / L. Conclusion Puerarin can promote the proliferation of MCF-7 cells in a dose-dependent manner, suggesting weak pheromone-like effect.
