目的 探讨乙酰肝素酶反义寡核苷酸对裸鼠食管癌移植瘤组织中uPA蛋白表达的影响。方法 采用免疫组化SP法对裸鼠食管癌移植瘤组织中uPA蛋白的表达进行了检测。结果 4条乙酰肝素酶反义寡核苷酸 (2 0mol/L)对裸鼠体内食管癌移植瘤中uPA蛋白表达表现出不同程度的抑制作用 ,实验组中uPA蛋白表达与N -ODN转染组、未经转染的空白对照组相比 ,差异均有统计学意义 (P <0 .0 5 )。结论 乙酰肝素酶反义寡核苷酸通过对裸鼠食管癌移植瘤组织中uPA蛋白表达 ,从而抑制肿瘤的浸润转移。
Objective To investigate the effect of heparanase antisense oligonucleotides on the expression of uPA protein in esophageal cancer xenografts in nude mice. Methods The expression of uPA protein in esophageal cancer xenografts in nude mice was detected by immunohistochemical SP method. Results Four heparanase antisense oligodeoxynucleotides (20 mol / L) inhibited the expression of uPA protein in esophageal cancer xenografts in nude mice to varying degrees. In the experimental group, uPA protein expression was positively correlated with N-ODN turnover Compared with blank control group, untransfected control group, the differences were statistically significant (P <0.05). Conclusion Heparanase antisense oligonucleotide inhibits the invasion and metastasis of tumor through the expression of uPA protein in the transplanted esophageal cancer tissue of nude mice.
