目的:优选黄芩的浸渍-压榨提取工艺。方法:采用浸渍-压榨联合提取法,选取提取时间、提取温度、溶剂倍数为考察因素,以总黄酮、黄芩苷提取率和出膏率的总评归一值(OD)为指标,通过Box-Behnken试验设计优选提取工艺。结果:最佳浸渍-压榨提取工艺为加11倍量水于60℃浸泡4 h,压榨,药渣加2倍量水洗涤后压榨,重复操作1次。总黄酮、黄芩苷提取率分别为17.630%,9.38%,出膏率42.7%,与模型预测值非常接近,说明该模型比较可靠。结论:优选的提取工艺科学、合理、可行,为中药常温提取工艺推广提供参考。
OBJECTIVE: To optimize the digestion-extraction process of Scutellaria baicalensis Georgi. Methods: The total extraction value (OD) of total flavonoids, baicalin and the rate of ointment were determined by the combined method of dipping and squeezing, extraction time, extraction temperature and solvent multiples. Experimental design optimization of extraction process. Results: The best impregnation - extraction process for the extraction of 11 times the amount of water soaked at 60 ℃ 4 h, squeezing, dregs plus 2 times the amount of water after washing press operation repeated 1 time. The extraction rates of total flavonoids and baicalin were 17.630%, 9.38% and 42.7%, respectively, which were very close to those predicted by the model, indicating that the model is more reliable. Conclusion: The optimal extraction process is scientific, reasonable and feasible, which can provide a reference for the generalization of the extraction process of Chinese herbal medicine at room temperature.
