目的 :建立测定核桃楸叶中槲皮素与山奈酚的高效液相色谱方法。方法:采用色谱柱DiamonsilC18(4.6mm×250mm,5μm);流动相:甲醇-0.2%磷酸(57:43),流速1.0mL/min,检测波长360nm,柱温:35℃。结果:槲皮素、山奈酚与其他组分的色谱峰基线分离,分离度均>1.5;槲皮素加样回收率平均值为98.62%,RSD为1.45%,山奈酚加样回收率为99.02%,RSD为1.11%。结论:该方法样品处理简单,准确度高,分离度好,重现性好,可用于核桃楸叶的质量控制。
Objective: To establish a HPLC method for the determination of quercetin and kaempferol in Juglans mandshurica leaves. METHODS: Diamonsil C18 column (4.6 mm × 250 mm, 5 μm) was used as the mobile phase. The mobile phase consisted of methanol-0.2% phosphoric acid (57:43), flow rate 1.0 mL / min, detection wavelength 360 nm and column temperature 35 ℃. Results: The chromatographic peaks of quercetin, kaempferol and other components were separated by baseline with the separation> 1.5. The average recovery of quercetin was 98.62%, RSD was 1.45%, and the recovery of kaempferol was 99.02 %, RSD is 1.11%. Conclusion: The method has the advantages of simple processing, high accuracy, good resolution and good reproducibility, and can be used for quality control of Juglans mandshurica leaf.
