目的:采用将集落法和MTT法结合发展而成的双琼脂MTT法进行肿瘤药敏试验,以克服MTT法中正常细胞干扰测定的缺点。方法:对小鼠肿瘤细胞及正常细胞株用两种方法的光密度、抑制率进行比较。结果:正常细胞在用双琼脂MTT法时生长不良,而肿瘤细胞则生长良好。结论:双琼脂MTT法优于MTT法。
OBJECTIVE: To use the double agar MTT method developed by combining the colony method and MTT method to carry out tumor susceptibility test to overcome the shortcomings of normal cell interference measurement in MTT assay. Methods: The optical density and inhibition rate of mouse tumor cells and normal cell lines were compared by two methods. Results: Normal cells grew poorly when double-agar MTT assay was used, while tumor cells grew well. Conclusion: The double agar MTT method is superior to the MTT method.
