目的:观察不同浓度尿酸盐溶液对人血管内皮细胞(Endothelial cell of vessels,ECV)分泌肿瘤坏死因子-α(TNF-α)的影响。方法:体外培养人血管内皮细胞,分别用低、中、高(710μmol/L、1070μmol/L、1430μmol/L)等不同浓度的尿酸盐刺激,用ELISA技术观测尿酸盐对血管内皮细胞分泌TNF-α的影响。结果:低、中、高浓度尿酸盐刺激组细胞上清液中TNF-α浓度均较空白对照组明显上调,均具有显著性统计学差异(P均<0.05)。结论:说明尿酸盐能直接作用于人的血管内皮细胞,诱导细胞分泌肿瘤坏死因子-α(TNF-α),这也可能是其促进动脉粥样硬化斑块形成的重要机制之一。
Objective: To observe the effects of different concentrations of urate solution on the secretion of tumor necrosis factor-α (TNF-α) by human endothelial cells of vessels (ECV). Methods: Human umbilical vein endothelial cells were cultured in vitro and stimulated with urate at different concentrations (710μmol / L, 1070μmol / L, 1430μmol / L), respectively. The effects of urate on the secretion of vascular endothelial cells TNF-α impact. Results: The concentrations of TNF-α in the supernatant of urate-stimulated group were significantly higher than those in the blank control group, both of which were significantly different (all P <0.05). CONCLUSION: Urate can directly act on human vascular endothelial cells and induce tumor cells to secrete tumor necrosis factor-α (TNF-α), which may also be one of its important mechanisms to promote the formation of atherosclerotic plaque.
