目的 研究慢性束缚应激时大鼠行为学变化、皮层和海马CA1区神经营养蛋白 3(NT3)的变化以及逍遥散、四君子汤、金匮肾气丸对其的影响。方法 用特制束缚架连续束缚 7d与 2 1d、每天 3h的方法制作大鼠束缚应激模型 ,用免疫组织化学方法结合图像分析检测大鼠皮层和海马CA1区NT3的变化。结果 2 1d模型组大鼠中央格停留时间明显延长 ,穿格次数明显减少 ,7d模型组大鼠的穿格数减少 ;两个模型组大鼠的挣扎次数明显减少。 3个复方都能缩短中央格停留时间 ,增加穿格次数和挣扎次数。连续束缚 7d后大鼠大脑皮层的NT3开始下降 ,至 2 1d模型组与正常对照组相比均有显著下降 (P <0 0 5 )。连续束缚 7d、2 1d后大鼠海马CA1区的NT3与正常对照组相比积分光密度明显降低 (均P <0 0 5 )。 3个复方均能升高皮层和海马NT3的积分光密度和海马中NT3的阳性细胞数 ,逍遥散能升高皮层NT3的阳性细胞数。结论 皮层和海马CA1区NT3下降参与了慢性应激的变化 ,疏肝、健脾、补肾的中药复方均能改善大鼠的行为变化 ,能上调NT3;但以逍遥散的调节作用较强 ,优于四君子汤和金匮肾气丸
Objective To study the changes of behavior, changes of neurotrophin 3 (NT3) in cortex and hippocampal CA1 region, and the effects of Xiaoyao San, Sijunzi Decoction and Jinkui Shenqi Pills on chronic restraint stress. Methods Rat restraint stress model was established by using a special restraint frame for 7 days and 21 days and 3 hours per day. Immunohistochemistry and image analysis were used to detect changes in rat cortex and hippocampal CA1 NT3. Results The dwelling time in the central lattice of rats in the 1d model group was significantly prolonged, the number of passages was significantly reduced, and the number of rats in the 7d model group was decreased. The number of struggles in the two model rats was significantly reduced. All three compounds can shorten the central lattice residence time and increase the number of times and struggles. After continuous restraint for 7 days, the NT3 in the rat cerebral cortex began to decline, and it was significantly decreased in the model group compared with the normal control group on the 21st day (P<0 05). The integrated optical density of NT3 in hippocampal CA1 region of rats was significantly lower than that of normal control group after continuous restraint for 7 days and 21 days (all P < 0.05). All three compounds can increase the integrated optical density of NT3 in the cortex and hippocampus and the number of NT3 positive cells in the hippocampus. Xiaoyao Powder can increase the number of NT3 positive cells in the cortex. Conclusions The decrease of NT3 in cortical and hippocampal CA1 regions is involved in the changes of chronic stress. The Chinese herbal compound of soothing the liver, spleen and tonifying the kidney can improve the behavioral changes of rats and can upregulate NT3; however, the regulation of Xiaoyao San is strong and excellent.
