应用SPC—A1人肺腺癌细胞系研究了顺铂和卡铂的细胞毒效应及其与人外周血LILAK细胞的联合效应及机制。结果表明:经一定浓度的顺铂或卡铂作用一定时间后,SPC—A1细胞可受到不同程度的杀伤。经1vg/ml的顺铂处理24小时,洗去药物后加入LILK细胞,SPC—A1细胞被明显杀伤,与未经顺铂处理的对照组相比P<0.001;而卡铂在同样条件下却无此效应。由于经顺铂处理的SPC—A1细胞对TNF—2有杀伤敏感性明显增加,推测该因子可能是介导顺铂与LILAK细胞协同效应的机制之一。
The SPC-A1 human lung adenocarcinoma cell line was used to investigate the cytotoxic effects of cisplatin and carboplatin and their combined effects with human peripheral blood LILAK cells and their mechanisms. The results showed that after a certain concentration of cisplatin or carboplatin for a certain period of time, SPC-A1 cells can be killed to varying degrees. After treatment with 1 vg/ml cisplatin for 24 hours, LILK cells were added after washing the drug, SPC-A1 cells were significantly killed, and P<0.001 compared with the control group without cisplatin treatment; while carboplatin was in the same condition. There is no such effect. Since cisplatin-treated SPC-A1 cells have a markedly increased cytotoxicity against TNF-2, it is speculated that this factor may be one of the mechanisms that mediate the synergistic effect of cisplatin and LILAK cells.
