仙蓟化斑胶囊抗变态反应的作用机制及其对皮肤过敏反应、肥大细胞脱颗粒释放的抑制功效

目的针对仙蓟化斑胶囊抗变态反应的作用机制及其对皮肤过敏反应(PCA)、肥大细胞脱颗粒释放的抑制功效进行研究。方法选择130只大鼠作为研究的对象,取10只用于制备抗卵蛋白血清,其余120只随机均分为实验一组(抗卵蛋白血清皮内注射,PCA实验)、二组(抗卵蛋白血清皮下注射,颅骨骨膜肥大细胞脱颗粒实验)、三组(抗卵蛋白血清腹腔注射,肥大细胞释放组胺实验),各组均含对照组、高剂量观察组、低剂量观察组、阳性药物组四个亚组,每组10只大鼠。采用灌胃方式,对照组给予生理盐水,高、低剂量观察组分别给予仙蓟化斑胶囊1.45 g/kg、0.70 g/kg,阳性药物组给予息斯敏0.0009 g/kg。比较分析各实验中,大鼠的PCA、肥大细胞释放组胺、骨膜肥大细胞脱颗粒等指标在不同处理下(高、低剂量仙蓟化斑胶囊,息斯敏,生理盐水)的差异。结果在注射同一浓度血清时,高剂量、低剂量观察组和阳性药物组大鼠的皮斑直径均小于对照组大鼠(P均<0.05);但三个给药组之间皮斑直径无差异(P>0.05)。与对照组比较,高剂量、低剂量观察组和阳性药物组大鼠的骨膜肥大细胞脱颗粒比例均下降(P均<0.01);且按低剂量观察组、阳性药物组和高剂量观察组之序递降。与对照组比较,高剂量、低剂量观察组和阳性药物组大鼠的肥大细胞释放组胺OD值均下降(P均<0.01);且按低剂量观察组、阳性药物组和高剂量观察组之序递降。结论仙蓟化斑胶囊可缩小大鼠PCA的皮斑直径,抑制肥大细胞脱颗粒及组胺释放,具有抗变态反应的作用。 OBJECTIVE To study the anti-allergic mechanism of Xianji Huaze Capsule and its inhibitory effect on skin allergic reaction (PCA) and degranulation of mast cells. Compared with the control group, the OD value of histamine released by the mast cells in the high-dose, low-dose observation group and the positive drug group decreased (all P <0.01); and by the low-dose observation group, the positive medicine group and the high-dose observation group Descending order. Conclusion Xianji Huaze Capsule can reduce the skin patch diameter of rat PCA, inhibit mast cell degranulation and histamine release, with anti-allergic effects.
