目的:研究大黄素与DNA相互作用。方法:通过共振光散射光谱和荧光光谱,发现大黄素与DNA相互作用产生强烈增强的共振光散射和荧光信号。结果:DNA与大黄素作用在556nm处产生较强共振散射峰,在540nm有很强的荧光峰,且荧光和散射强度均呈良好的线性关系。结论:通过Scatchard方程和双倒数曲线得出大黄素与DNA的结合常数是KA=0.18×105L·mol-1,说明它能嵌入到DNA碱基对之间形成复合物,也是一种DNA嵌入剂。
Objective: To study the interaction between emodin and DNA. METHODS: Through resonance light scattering and fluorescence spectroscopy, it was found that emodin interacts with DNA to produce strongly enhanced resonance light scattering and fluorescence signals. Results: The interaction between DNA and emodin produced a strong resonance scattering peak at 556 nm, a strong fluorescence peak at 540 nm, and a good linear relationship between fluorescence and scattering intensity. Conclusion: The binding constant of emodin to DNA is determined by the Scatchard equation and the double reciprocal curve to be KA=0.18×105L·mol-1, indicating that it can be embedded in the DNA base pairs to form a complex, which is also a DNA intercalator .
