以黄花柳花为试材,采用紫外分光光度法,选择5种不同型号的树脂进行静态吸附试验,筛选出对其吸附解吸效性能高的树脂,再通过动态吸附与解吸试验研究上样浓度、树脂上样量、洗脱剂及流速等条件对黄花柳花总黄酮纯化效果的影响,优化维药黄花柳花总黄酮的大孔树脂纯化工艺。结果表明:AB-8型大孔吸附树脂对黄花柳花总黄酮具有较好的吸附解吸效果,最佳纯化条件为黄花柳花待纯化液浓度为3.13g/L,调节上样液pH 4.2,上样量5.5mL/g,以1.0mL/min流速上样,依次用10BV蒸馏水淋洗,6BV 70%乙醇以2.0mL/min流速洗脱,得纯化液。经AB-8纯化后的黄花柳花总黄酮纯度达51.99%。AB-8型大孔吸附树脂能有效提高黄花柳花中总黄酮纯度,并符合中药有效部位研究要求。
Using the method of UV spectrophotometry, five different types of resin were selected for the static adsorption test, the resin with high performance of adsorption and desorption was screened out, and the concentration of the sample was studied by dynamic adsorption and desorption test. The results showed that AB-8 macroporous resin had good adsorption and desorption effects on the total flavonoids of Huanghua Liuhua. The optimal purification conditions were as follows: the concentration of the purified Huanghua Liuhua was 3.13g / L, the pH of the sample was adjusted to 4.2, The sample volume was 5.5mL / g, and the sample was loaded at a flow rate of 1.0mL / min. The sample was washed with 10BV distilled water and then with 6BV 70% ethanol at a flow rate of 2.0mL / min. After purified by AB-8, the purity of total flavonoids of S. aureus reached 51.99%. AB-8 macroporous resin can effectively improve the purity of the total flavonoids in the flower of the flower and meet the research requirements of effective parts of traditional Chinese medicine.
