目的:探讨凋亡抑制蛋白(inhibitor of apoptosis protein,IAP)家族的新成员Livinα和Livinβ及其基因在人口腔鳞癌组织中的表达及意义。方法:采用反转录聚合酶链反应法(RT-PCR)检测20例口腔鳞癌组织、15例口腔软组织囊肿和10例癌旁正常口腔黏膜组织中LivinαmRNA和LivinβmRNA的表达,并用Western印迹检测2种Livin蛋白在3种组织中的表达情况,应用SPSS10.0软件包对结果进行统计学分析。结果:19例口腔鳞癌组织中均有Livinα和LivinβmRNA的表达,14例口腔软组织囊肿和10例癌旁正常口腔黏膜组织中均可见Livinα或LivinβmRNA的表达,3者表达率无统计学差异,但Livinα和LivinβmRNA在口腔鳞癌组织中的表达显著高于口腔软组织囊肿和癌旁正常口腔黏膜组织(P<0.01),Western印迹检测结果和RT-PCR结果一致。结论:Livin可能在人口腔鳞癌的发生、发展中起着重要作用。
Objective: To investigate the expression and significance of Livinα and Livinβ, a novel member of the inhibitor of apoptosis protein (IAP) family in human oral squamous cell carcinoma. Methods: The expressions of LivinαmRNA and LivinβmRNA were detected by reverse transcription polymerase chain reaction (RT-PCR) in 20 cases of oral squamous cell carcinoma, 15 cases of oral soft tissue cysts and 10 cases of normal para-cancerous mucosa tissues. The expression of LivinαmRNA and LivinβmRNA were detected by Western blot The expression of Livin protein in three kinds of tissues was analyzed by SPSS10.0 software package. Results: The expression of Livinα and LivinβmRNA in all the 19 cases of oral squamous cell carcinoma were detected. The expression of Livinα or LivinβmRNA was found in 14 cases of oral soft tissue cyst and 10 cases of adjacent normal mucosa. There was no significant difference among the three groups The expression of Livinαand LivinβmRNA in oral squamous cell carcinoma tissues was significantly higher than that in oral soft tissue cysts and adjacent normal mucosa tissues (P <0.01). Western blot results were consistent with RT-PCR results. Conclusion: Livin may play an important role in the occurrence and development of oral squamous cell carcinoma.
