原子力显微镜对骨髓CD34+细胞的膜表面超微结构研究

本研究目的是利用原子力显微镜 (atomicforcemicroscope ,AFM)观察骨髓CD3 4 +造血细胞的膜表面超微结构 ,对比正常人与白血病患者CD3 4 +造血细胞膜表面超微结构的形态学差异。将免疫磁珠分离仪富集的CD3 4 +细胞滴至新鲜剥离的云母片上 ,在空气中干燥后用AFM进行观察。结果表明 :利用AFM附带软件IP2 .1的线分析及面分析功能 ,得到正常人与白血病患者CD3 4 +造血细胞膜表面结构的几何参数 ,经比较 ,白血病患者CD3 4 +造血细胞的高低差Rp v、均方根粗糙度Rrms、平均粗糙度Ra、平均高度 Z 4个几何参数值均明显大于正常人。结论 :AFM能准确直观地观察到CD3 4 +造血细胞的膜表面超微结构 ,并同时获得其膜表面特征几何参数 ,可为临床白血病的快速诊断、HSPCT移植物筛选和预后提供重要参考。 The purpose of this study was to observe the ultrastructure of the membrane surface of CD3 + hematopoietic cells by atomic force microscopy (AFM) and to compare the morphological differences of the ultrastructure of the membrane surface of CD34 + hematopoietic cells in normal and leukemic patients. CD3 4 + cells enriched in the immunomagnetic bead separator were dropped on freshly peeled mica flakes, dried in air and observed with AFM. The results showed that the geometrical parameters of the membrane surface structure of CD34 + hematopoietic cells in normal people and leukemia patients were obtained by the line analysis and surface analysis function of the attached software IP2.1 of AFM. By comparison, the difference of Rp v , Root mean square roughness Rrms, average roughness Ra, average height Z four geometric parameters were significantly greater than normal. CONCLUSION: AFM can accurately and directly observe the membrane surface ultrastructure of CD3 4 + hematopoietic cells and obtain the geometrical parameters of the membrane surface at the same time. It can provide an important reference for the rapid diagnosis of clinical leukemia and the screening and prognosis of HSPCT.
