目的:观察四味降脂汤对肥胖大鼠血清神经肽Y(NPY)含量和下丘脑NPY表达的影响,探讨四味降脂汤减肥的机制。方法:用高脂饲料喂养制作肥胖大鼠模型,将造模成功的实验性肥胖大鼠随机分为模型组和中药组加以治疗。放射免疫法检测血清NPY含量,免疫组织化学法检测NPY表达。结果:四味降脂汤组血清NPY含量和下丘脑NPY表达比模型组降低(P<0.01);血清NPY水平与普食组比较仍有一定的差异(P<0.01)。结论:肥胖大鼠NPY含量的改善是四味降脂汤减肥的作用机制之一。
Objective: To observe the effect of Siwei Jiangzhi Decoction on the content of neuropeptide Y (NPY) and the expression of NPY in hypothalamus of obese rats, and to explore the mechanism of reducing weight of Siwei Jiangzhi decoction. METHODS: Obese rat models were fed with high-fat diet and experimentally obese rats with successful modeling were randomly divided into model group and Chinese medicine group. Serum NPY levels were measured by radioimmunoassay and NPY expression was detected by immunohistochemistry. Results: The serum NPY content and hypothalamic NPY expression in Siwei Jiangzhi decoction group were lower than that in the model group (P<0.01). The serum NPY level was still different from that in the normal food group (P<0.01). Conclusion: The improvement of NPY content in obese rat is one of the mechanisms of weight loss of Siwei Jiangzhi decoction.
