目的 探索 5 ,10 亚甲基四氢叶酸还原酶 (MTHFR)基因与血压水平的连锁关系。方法 研究现场选择在原发性高血压社区综合防治项目农村社区 -北京市房山区良各庄村和黄山店村。研究对象为该现场的 142对同胞对 ,及可能获得的父母。基因型测量包括血凝块中提取DNA ,PCR扩增目的基因片断 ,酶切分析MTHFR基因多态性。表型为血压水平 ,协变量的测量包括问卷调查、体格指标及血液生化指标的测量。统计分析方法为数量型同胞对连锁分析 ,分析软件为SAGE。结果 在研究人群MTHFR基因 6 77位C和T等位基因频率分别为 0 .70和 0 .30 ,基因型频率 (CC、CT、TT)分别为 49.7%、41.0 %和 9.3%。没有发现MTHFR基因与血压水平存在连锁 (P >0 .0 5 )。结论 本研究人群MTHFR基因不太可能是决定血压水平的主基因。
Objective To explore the linkage between 5, 10 methylenetetrahydrofolate reductase (MTHFR) gene and blood pressure level. Methods The study site was selected in the rural community of integrated community hypertension prevention project - Lianggezhuang Village and Huangshan Village in Fangshan District, Beijing. The object of study was 142 pairs of fellow patriots on the site, and possible parents. Genotyping included DNA extraction from blood clots, PCR amplification of the target gene fragment, and restriction analysis of the MTHFR gene polymorphism. Phenotypes were blood pressure levels, and covariates were measured by questionnaires, physical markers, and blood biochemical markers. Statistical analysis method for quantitative sibling pair analysis, analysis software for SAGE. Results The prevalence of C allele and C allele at 677th locus in MTHFR gene were 0.70 and 0.30, respectively. The genotype frequencies (CC, CT, TT) were 49.7%, 41.0% and 9.3% respectively. No MTHFR gene was found to be linked to blood pressure (P> 0.05). Conclusions The MTHFR gene in this study population is unlikely to be the major gene that determines blood pressure.
