目的研究Ang-1、Ang-2在鼻咽癌组织中的表达情况与鼻咽癌淋巴管密度的相关性。方法用免疫组化法检测Ang-1、Ang-2在鼻咽癌组织及鼻咽部慢性炎症中的表达情况,用单克隆抗体D2-4 0标记鼻咽癌瘤内及瘤周微淋巴管。分析Ang-1、Ang-2与鼻咽癌临床病理参数与微淋巴管密度之间的关系。结果 Ang-1、Ang-2在鼻咽癌组织中阳性表达率分别为46.15%(24/52)、63.5%(33/5 2),明显高于鼻咽部炎性组织2 5.0%(3/1 2)、3 3.3%(4/1 2)(P<0.0 5)。Ang-1在鼻咽癌组织中的表达与性别、年龄、T分级及临床分期、有无淋巴结转移等因素均无明显差异;Ang-2在鼻咽癌组织中的表达与性别、年龄、T分级及临床分期等因素无明显差异,但在淋巴结转移组中的表达率83.9%(26/3 1)明显高于无淋巴结转移组3 3.3%(7/2 1)(P<0.0 1)。D2-4 0标记鼻咽癌微淋巴管阳性表达率达9 2.3 1%(4 8/5 2),并且在淋巴结转移组中瘤内及瘤周微淋巴管密度均高于无淋巴结转移组(P<0.0 5)。联合Ang-1、Ang-2在鼻咽癌组织中的表达与瘤内及瘤周微淋巴管密度均成正相关。结论Ang-2与鼻咽癌淋巴结转移密切相关,微淋巴管密度可作为判断淋巴结有无转移的重要指标之一。
Objective To investigate the correlation between the expression of Ang-1 and Ang-2 in nasopharyngeal carcinoma and lymphatic vessel density of nasopharyngeal carcinoma. Methods The expression of Ang-1 and Ang-2 in nasopharyngeal carcinoma and chronic inflammation of nasopharyngeal tissues were detected by immunohistochemistry. The intra-nasal and peritumor lymphatic vessels were labeled with monoclonal antibody D2-4 0 . Ang-1, Ang-2 and nasopharyngeal carcinoma clinicopathological parameters and the relationship between the lymphatic vessel density. Results The positive rates of Ang-1 and Ang-2 in nasopharyngeal carcinoma tissues were 46.15% (24/52) and 63.5% (33/5 2), respectively, which were significantly higher than those in nasopharyngeal tissues (5.0%, 3 / 1 2), 3 3.3% (4/1 2) (P <0.0 5). Ang-1 expression in nasopharyngeal carcinoma and gender, age, T grade and clinical stage, with or without lymph node metastasis and other factors were not significantly different; Ang-2 expression in nasopharyngeal carcinoma and gender, age, T There was no significant difference between the two groups (P <0.01). However, the expression rate in lymph node metastasis group was 83.9% (26/3 1) which was significantly higher than 3.3% (7/2 1) in non-lymph node metastasis group (P <0.01). The positive rate of D2-4 0 labeling in lymphatic vessels of nasopharyngeal carcinoma was 92.3% (48/52), and the lymphatic vessel density in the lymph node metastasis group was higher than that in the non-lymphatic metastasis group P <0.0 5). The expression of Ang-1 and Ang-2 in nasopharyngeal carcinoma was positively correlated with the lymphatic vessel density in the tumor and peritumoral. Conclusions Ang-2 is closely related to lymph node metastasis of nasopharyngeal carcinoma, and lymphatic vessel density can be used as one of the important indexes to judge whether there is lymph node metastasis.
