[目的]探讨脂蛋白脂酶(lipoprotein lipase,LPL)基因PvuⅡ位点多态性与原发性高血压合并肥胖的相关性。[方法]采用聚合酶链反应和限制性酶切片段长度多态性(PCR-RFLP)技术,对135名原发性高血压合并肥胖患者和135名健康对照LPL基因PvuⅡ酶切位点进行多态性分析。[结果]LPL基因PvuⅡ位点P+P+,P+P-和P-P-3种基因型的构成比分别为36.7%、45.6%和17.8%。等位基因的频率为59.4%(P+)和40.6%(P-)。PvuⅡ位点基因型分布在病例组与对照组间有统计学差异(P﹤0.01),其中P-P-的WHR低于P+P-和P+P+基因型,而DBP水平高于其他两种基因型(P值均﹤0.01)。[结论]LPL基因PvuⅡ位点多态性与宁夏银川市农村人群EH合并肥胖的发病有关,其中PvuⅡ位点(-/-)基因型与WHR的降低和DBP水平的升高有关。
[Objective] To investigate the relationship between polymorphism of Pvu Ⅱ locus in lipoprotein lipase (LPL) gene and essential hypertension complicated with obesity. [Methods] PCR-RFLP technique was used to detect Pvu Ⅱ cleavage sites of LPL gene in 135 patients with essential hypertension complicated with obesity and 135 healthy controls Analysis of the state. [Result] The proportions of P + P +, P + P- and P-P-3 genotypes in the PvuⅡ locus of LPL gene were 36.7%, 45.6% and 17.8% respectively. The frequency of alleles was 59.4% (P +) and 40.6% (P-). The genotype distribution of PvuⅡwere statistically different between the case group and the control group (P <0.01). The WHR of PP- was lower than that of P + P- and P + P + genotypes, while the level of DBP was higher than that of the other two genes Type (all P <0.01). [Conclusion] The polymorphism of Pvu Ⅱ locus in LPL gene is associated with the incidence of EH complicated with obesity in Ningxia Yinchuan. The genotype of Pvu Ⅱ locus (- / -) is related to the decrease of WHR and the increase of DBP level.
