目的 了解海南省恶性疟原虫裂殖子表面蛋白 1(pfMSP1)和表面蛋白 2 (pfMSP2 )等位基因型的分型特征。方法 采用套式PCR法分别扩增pfMSP1基因第 2区与第 3区部分片段和pfMSP2基因的中间可变区 (第 3区 ) ,对海南省恶性疟原虫分离株进行基因分型。结果 5 5份血样中有 5 2份恶性疟疾患者扩增出pfMSP1基因片段 ,以MAD2 0型为主导型 (84 6 % ) ,K1型为次要型 ,未检出RO33型 ,两种不同等位基因混合感染率为 15 4 %。同时 ,5 5份血样中有 5 3份恶性疟疾患者扩增出pfMSP2基因片段 ,以 3D7型为主导型 (83% ) ,FC2 7型为次要型 ,混合感染率为 17%。结论 海南省恶性疟原虫的MSP1存在MAD2 0型和K1型两种等位基因型 ,以MAD2 0型为优势虫株 ;其MSP2存在 3D7型和FC2 7型两种等位基因型 ,以 3D7等位基因家族为主。
Objective To understand the typing characteristics of Plasmodium falciparum surface protein 1 (pfMSP1) and surface protein 2 (pfMSP2) alleles in Hainan Province. Methods The nested variable region (Region 3) of partial region 2 and region 3 of pfMSP1 gene and the pfMSP2 gene were amplified by nested PCR. The genotypes of Plasmodium falciparum isolates from Hainan Province were genotyped. Results The pfMSP1 gene fragment was amplified from 52 samples of 5 malaria patients, accounting for MAD20 type (84.6%), K1 type was minor type, RO33 type was not detected, two different The bit-mixed rate was 15 4%. At the same time, pfMSP2 gene fragment was amplified in 53 blood samples from 5 malaria patients. The 3D7-type was predominant (83%), while the FC27 was secondary, with a mixed infection rate of 17%. Conclusion MSP1 of Plasmodium falciparum in Hainan Province has two genotypes of MAD2 0 and K1, and MAD2 0 is the dominant strain. There are two alleles of MSP2 in 3D7 and FC27, The main gene family.
