目的:探讨六味地黄丸中马钱苷、芍药苷、莫诺苷、丹皮酚4种入血成分组合对大鼠前脂肪细胞增殖、分化的影响,并优选其最佳浓度配伍。方法:原代培养大鼠的前脂肪细胞,将马钱苷等4种入血成分按四因素三水平的正交试验表L9(34)安排实验,同时设立空白对照组,以四甲基偶氮唑盐(MTT)法测定细胞增殖的速度,以油红O染色方法检测其对大鼠前脂肪细胞分化过程中的细胞内脂肪积聚的影响,以光密度为指标比较各因素、水平之间的差异。结果:六味地黄丸4种入血成分组合对大鼠前脂肪细胞增殖、分化的影响以组合A3B1C3D2的作用最为显著(P<0.01);与各单体组分作用比较,具有高度显著性差异(P<0.01)。结论:六味地黄丸4种入血成分以马钱苷、莫诺苷高浓度,丹皮酚中浓度,芍药苷低浓度配伍组合对大鼠前脂肪细胞增殖、分化影响较单体组分作用显著,表现为中药组方的整体性和协同增效调节作用。
OBJECTIVE: To investigate the effects of four infusions of loganin, paeoniflorin, morroniside and paeonol on the proliferation and differentiation of rat preadipocytes in Liuweidihuang Pills, and to optimize the optimal combination of them. Methods: Primary rat preadipocytes were cultured, and the four infusions of loganin were arranged in an orthogonal test table L9 (34) of four factors and three levels. Results: The effects of the four components of Liuweidihuang Pills on the proliferation and differentiation of preadipocytes in combination with A3B1C3D2 were the most significant (P <0.01). The effects of four components of Liuweidihuang Pills on the proliferation and differentiation of rat preadipocytes were highly significant (P <0.01) P <0.01). CONCLUSION: The four components of Liuweidihuangwan can significantly affect the proliferation and differentiation of rat preadipocytes compared with the single component in the combination of loganin, high concentration of morroniside, middle concentration of paeonol and low concentration of paeoniflorin , Manifested as a unit of traditional Chinese medicine group and synergies regulation.
