目的:利用RNA干涉技术抑制肿瘤细胞端粒酶表达,探讨其对肿瘤细胞凋亡的诱导作用。方法:将成功构建的针对hTERT的siRNA(smallinterferencingRNA)表达载体转染HeLa细胞,通过透射电镜、免疫印迹和流式细胞术(FCM)等方法检测人宫颈癌HeLa细胞的凋亡情况。结果:构建的siRNA表达载体可以诱导HeLa细胞发生凋亡。结论:针对人端粒酶逆转录酶的siRNA表达载体稳定转染HeLa细胞,可诱导HeLa细胞发生凋亡。
OBJECTIVE: To inhibit the expression of telomerase in tumor cells by RNA interference and to explore the induction of apoptosis on tumor cells. Methods: HeLa cells were transfected with siRNA targeting small interfering RNA (shRNA) targeting hTERT. The apoptosis of human cervical cancer HeLa cells was detected by transmission electron microscopy, Western blotting and flow cytometry (FCM). Results: The constructed siRNA expression vector could induce HeLa cell apoptosis. Conclusion: HeLa cells transfected with siRNA targeting human telomerase reverse transcriptase can induce apoptosis in HeLa cells.
