目的探讨食管癌中 p75~(NTR)阳性细胞池的变化与顺铂耐药间的关系。方法采用流式细胞技术检测未处理组和顺铂作用组的不同时间点5株食管癌细胞中 p75~(NTR)阳性细胞池的变化情况。随后,用免疫磁珠方法将食管癌细胞分为 p75~(NTR)阳性细胞池和阴性细胞池,观察 p75~(NTR)阳性细胞池的分化情况、检测顺铂作用2 d 后食管癌细胞的凋亡率。结果未加药的食管癌细胞中p75~(NTR)阳性细胞池的比例一直维持恒定,但是在顺铂作用下,5株食管癌细胞中 p75~(NTR)阳性细胞池的比例均有明显上升。在分化实验中可观察到 p75~(NTR)阳性细胞可生成 p75~(NTR)阳性和阴性细胞;顺铂作用下,p75~(NTR)阳性细胞池的凋亡率明显低于 p75~(NTR)阴性细胞池。结论 p75~(NTR)阳性细胞池的自我分化和耐药性符合肿瘤干细胞的特征。
Objective To investigate the relationship between the change of p75 ~ (NTR) -positive cell pool and cisplatin resistance in esophageal cancer. Methods Flow cytometry was used to detect the changes of p75 ~ (NTR) positive cell pools in 5 esophageal cancer cells at different time points in untreated and cisplatin-treated groups. Subsequently, the esophageal cancer cells were divided into p75 ~ (NTR) positive cell pools and negative cell pools by immunomagnetic beads method. The differentiation of p75 ~ (NTR) positive cell pools was observed, and the effect of cisplatin on esophageal cancer cells Apoptosis rate. Results The proportion of p75 ~ (NTR) -positive cell pool in untreated esophageal cancer cells remained constant, but the proportion of p75 ~ (NTR) -positive cell pools in 5 esophageal cancer cells increased significantly under the action of cisplatin . P75 ~ (NTR) -positive cells were found to be positive for p75 ~ (NTR) and negative cells in differentiation experiment. The apoptotic rate of p75 ~ (NTR) positive cells was significantly lower than that of p75 ~ (NTR) ) Negative cell pool. Conclusion The self-differentiation and drug resistance of p75 ~ (NTR) -positive cell pool are consistent with the characteristics of tumor stem cells.
