目的观察孕鼠被动吸烟对胎鼠海马神经元一氧化氮合酶表达的影响,以期为探讨被动吸烟对子代危害的机制提供更多实验依据。方法将雌性大鼠随机分为对照组和实验组两组。实验组采用人工熏烟的方法建立被动吸烟模型,对照组不作处理。妊娠21日人工结束分娩,取胎鼠脑组织,进行一氧化氮合酶(NOS)免疫组化染色。高倍镜下对海马区观察并摄片,用图像分析软件Image-Pro Plus对两组切片进行光密度分析,观察其差别。结果 NOS在胎鼠海马区有表达,分布于海马神经元胞质内。计算机图像分析结果表明,与对照组比较,实验组海马NOS表达下降,统计结果有显著差异性(P<0.05)。结论孕鼠被动吸烟使胎鼠海马NOS的表达下降,其可能为孕期被动吸烟导致子代学习记忆记忆功能下降的机制之一。
Objective To observe the effects of passive smoking on the expression of nitric oxide synthase in fetal rat hippocampal neurons in order to provide more experimental evidence for exploring the mechanism of passive smoking harm to offspring. Methods Female rats were randomly divided into control group and experimental group. The experimental group established a passive smoking model by artificial fumigation, while the control group did not make any treatment. Pregnancy on the 21st artificial termination of childbirth, fetus brain tissue, nitric oxide synthase (NOS) immunohistochemical staining. The hippocampus was observed and photographed under high magnification. The optical density of the two groups of slices was analyzed by Image-Pro Plus software, and the difference was observed. Results NOS was expressed in fetal rat hippocampus and distributed in the cytoplasm of hippocampal neurons. The results of computer image analysis showed that compared with the control group, the expression of NOS in the hippocampus of the experimental group decreased and the statistical results were significantly different (P <0.05). Conclusion Passive smoking in pregnant rats decreases the expression of NOS in fetal rat hippocampus, which may be one of the mechanisms of passive smoking during pregnancy leading to the decline of learning and memory ability in offspring.
