本文应用单克隆抗体结合APAAP技术,Dot—blot地高辛标记核酸杂交技术,研究了哮喘患儿淋巴细胞CD23mRNA和膜CD23分子表达,同时检测了血清总IgE水平。结果显示:哮喘患者淋巴细胞CD23mRNA表达增高,而正常对照组未见杂交信号;外周血单个核细胞和B细胞CD23分子表达均明显增加,与对照组相比尸值分别<0.01,<0.001以B细胞CD23分子表达增加更显著。血清IgE水平异常升高,B细胞CD23分子表达与血清IgE水平之间呈显著正相关。结果提示CD23直接参与了过敏性哮喘的发病。
In this paper, monoclonal antibody combined with APAAP technology, Dot-blot digoxigenin-labeled nucleic acid hybridization technology to study the expression of CD23mRNA and membrane CD23 of lymphocytes in children with asthma, and to test the serum total IgE levels. The results showed that the expression of CD23 mRNA in lymphocytes was increased in asthmatic patients, while no signal was observed in the normal control group. CD23 expression in peripheral blood mononuclear cells and B cells were significantly increased as compared with the control group Cell CD23 molecule expression increased more significantly. Serum IgE levels were abnormally elevated, CD23 expression of B cells and serum IgE levels was significantly positively correlated. The results suggest that CD23 is directly involved in the pathogenesis of allergic asthma.
