目的:比较糖尿病性心肌病大鼠和中药复方糖心康治疗组大鼠心肌组织中基因表达的差异,并用Evegreen实时荧光RT-PCR技术定量检测胰岛素样生长因子结合蛋白-3(Igfbp-3mRNA)在各组大鼠心肌组织的表达水平。方法:(1)差异表达基因的筛选;(2)荧光定量RT-PCR的检测。结果:仅对筛选出差异表达基因谱中的Igfbp3mRNA进行报道。糖尿病性心肌病模型组心肌组织基因Igfbp3mRNA表达下调,中药复方肾消康治疗组心肌组织基因Ig-fbp3mRNA表达上调。结论:Igfbp-3mRNA在糖尿病状态下心肌组织也有表达,并且表达上调,而中药复方糖心康可使其下调,这也是中药复方糖心康对糖尿病性心肌组织损伤具有保护作用的机制之一。
OBJECTIVE: To compare the difference of gene expression in myocardial tissue of rats with diabetic cardiomyopathy and Chinese medicine compound Tangxinkang, and to quantitatively detect insulin-like growth factor binding protein-3 (Igfbp-3 mRNA) by Evegreen real-time fluorescence RT-PCR. The expression level of myocardial tissue in each group. Methods: (1) Screening of differentially expressed genes; (2) Detection of fluorescence quantitative RT-PCR. Results: Igfbp3 mRNA was screened only for the differentially expressed gene profiles. The expression of Igfbp3 mRNA in myocardium was down-regulated in the model group of diabetic cardiomyopathy, and the expression of Ig-fbp3 mRNA in myocardium tissue was up-regulated in the Chinese medicine compound Shenxiaokang group. Conclusion: Igfbp-3 mRNA is also expressed in diabetic myocardium and up-regulated in the myocardium. Diabetes can be down-regulated by traditional Chinese medicine compound Tangxinkang. This is also one of the protective mechanisms of Chinese herbal formula Tangxinkang on diabetic myocardial tissue injury.
