目的:研究前列腺素E1(PGE1)对糖尿病肾病(DN)大鼠肾组织细胞凋亡及相关蛋白Fas、bcl-2表达的影响并探讨其机理。方法:大鼠75只随机分为4组,前3组用链脲佐菌素(STZ)建立糖尿病肾病模型,并分别给予PGE1、洛汀新和生理盐水,第4组作为正常对照组。所有的大鼠在喂养101d后处死,取其肾脏,观察细胞凋亡数量及Fas和bc1-2表达。结果:PGE1治疗组治疗前后肾小球和肾小管凋亡细胞均较糖尿病组显著减少,具有统计学意义(P<0.05)。结论:PGE1可抑制糖尿病肾病大鼠肾组织细胞凋亡,下调Fas的表达、上调bcl-2的表达,其作用明显优于洛汀新。
Objective: To investigate the effect of prostaglandin E1 (PGE1) on the apoptosis of renal nephropathy and the expression of Fas and bcl-2 in diabetic nephropathy (DN) rats and its mechanism. Methods: Seventy five rats were randomly divided into four groups. The first three groups were given streptozotocin (STZ) to establish model of diabetic nephropathy. The rats were given PGE1, Lotensin and normal saline respectively. The fourth group served as the normal control group. All rats were killed after feeding for 101 days, their kidneys were taken out, the number of apoptosis and the expression of Fas and bc1-2 were observed. Results: The apoptosis of glomerular and tubular cells in PGE1 group was significantly lower than that in diabetic group before treatment (P <0.05). CONCLUSION: PGE1 can inhibit renal cell apoptosis, down-regulate the expression of Fas and up-regulate the expression of bcl-2 in diabetic nephropathy rats, and its effect is obviously superior to that of Lotensin.
