目的:研究柴胡皂苷d对大鼠肝星状细胞ERα、ERβmRNA的调节作用,探讨其对肝纤维化的作用机制。方法:体外培养大鼠肝星状细胞系HSC-T6,采用实时荧光定量RT-PCR方法,观察柴胡皂苷d对两种雌激素受体亚型表达的影响。采用MTT法,观察雌二醇、柴胡皂苷d对HSC-T6细胞增殖的影响,并为后续实验确定合适的实验用药物剂量和用药时间。结果:柴胡皂苷d能明显提高HSC-T6细胞ERα、βmRNA表达,且呈剂量与时间依赖性,但作用较雌二醇弱,这种作用可被ER完全拮抗剂ICI-182.780抑制。通过对半数抑制率的计算,得出柴胡皂苷d和雌二醇半数抑制率(IC50)的浓度分别为5μmol/l和1μmol/l。结论:推测柴胡皂苷d有雌激素受体调节剂的作用,可能是一种雌激素调节剂。
OBJECTIVE: To study the regulatory effect of saikosaponin-d on ERα and ERβmRNA in rat hepatic stellate cells and to explore its mechanism of action on hepatic fibrosis. Methods: Rat hepatic stellate cell line HSC-T6 was cultured in vitro. The effects of saikosaponin d on the expression of two estrogen receptor subtypes were observed by real-time fluorescence quantitative RT-PCR. The effects of estradiol and saikosaponin d on the proliferation of HSC-T6 cells were observed by MTT assay, and appropriate experimental dose and duration of drug were determined for subsequent experiments. Results: Saikoside S significantly increased the expression of ERα and β mRNA in HSC-T6 cells in a dose-and time-dependent manner, but its effect was weaker than that of estradiol. This effect was inhibited by ICI-182.780. Through the calculation of the half inhibition rate, the concentrations of saikosaponin d and estradiol (IC50) were 5μmol / l and 1μmol / l, respectively. Conclusion: It is speculated that saikosaponin d has estrogen receptor modulators and may be an estrogen modulator.
