本文应用未标记抗体过氧化物酶和抗过氧化物酶(PAP和双桥PAP)技术染色,保存5~20年,福尔马林固定的石蜡包埋标本,显示反应性增生和腺癌转移淋巴结中IgG产生细胞。分析比较减低背景着色的四种方法、PAP与双桥PAP法的染色效果,以及各种抗体和DAB液中H_2O_2浓度对染色效果的影响。进一步证实PAP法是适合在石蜡切片上进行回顾性研究的高敏感、高特异性方法。
In this paper, unlabeled antibody peroxidase and anti-peroxidase (PAP and double-bridged PAP) technique were used for staining and preserved for 5 to 20 years. Formalin-fixed paraffin-embedded specimens showed reactive hyperplasia and adenocarcinoma metastasis. IgG produces cells in lymph nodes. The four methods of reducing background coloration, the staining effects of PAP and double bridge PAP method, and the effects of H 2 O 2 concentration in various antibodies and DAB solutions on the staining results were analyzed and compared. It is further confirmed that the PAP method is a highly sensitive and highly specific method suitable for retrospective studies on paraffin sections.
