目的:观察银耳孢多糖对小鼠大肠癌的抑制作用,并对其机制进行初步探讨。方法:昆明种小鼠随机分为模型组和银耳孢多糖(高、中、低剂量)4组,小鼠右后肢背部皮下注射1×106个活CMT93细孢制备肿瘤病变模型,银耳孢多糖低、中、高剂量组分别给予银耳孢多糖5,10,20mg/kg.d灌胃每天1次,连续12天。取肿瘤瘤体,称瘤重,计算抑瘤率;分别采用realtime PCR、免疫组织化学方法检测肿瘤组织中VEGF-C mRNA和survivin、VEGF-C蛋白含量。结果:银耳孢多糖低剂量能明显降低肿瘤瘤重,降低肿瘤组织中VEGF-C mRNA和survivin、VEGF-C蛋白含量;中、高剂量组能显著降低肿瘤瘤重,降低肿瘤组织中VEGF-CmRNA和survivin、VEGF-C蛋白含量。结论:银耳孢多糖对小鼠大肠癌有明显抑制作用,其机制可能与降低survivin、VEGF-C表达有关。
OBJECTIVE: To observe the inhibitory effect of Tremella Polysaccharides on colorectal cancer in mice and to discuss its mechanism. METHODS: Kunming mice were randomly divided into model group and Tremella fusca polysaccharide (high, medium and low dose) groups. Mice were implanted with 1×106 live CMT93 spores subcutaneously on the back of the right hind limb to prepare a tumor lesion model. The high, medium and high dose groups were given intragastric spore polysaccharides 5, 10, 20 mg/kg.d once daily for 12 consecutive days. Tumorous tumors were harvested and tumor weights were calculated. Tumor inhibition rates were calculated; realtime PCR and immunohistochemistry were used to detect the VEGF-C mRNA, survivin, and VEGF-C protein contents in tumor tissues. RESULTS: Low dose of Tremella fusca polysaccharide could significantly reduce tumor weight and decrease the content of VEGF-C mRNA, survivin and VEGF-C protein in tumor tissue. The middle and high dose group could significantly reduce tumor weight and decrease VEGF-C mRNA in tumor tissue. And survivin, VEGF-C protein content. Conclusion: Tremella fusca polysaccharide can obviously inhibit the colorectal cancer in mice. The mechanism may be related to the decrease of survivin and VEGF-C expression.
