目的探讨雌激素受体2基因(ESR2)多态性与盐城地区青年胃腺癌的关联。方法应用聚合酶链反应-限制性片段长度多态性技术,对盐城地区100例青年胃腺癌患者(胃腺癌组)和100名青年胃溃疡患者(对照组)ESR2第5外显子RsaⅠ酶切多态和第8外显子AluⅠ酶切多态进行分析。结果(1)ESR2第8外显子AluⅠ酶切多态分析表明,胃腺癌组A等位基因频率明显高于对照组(P<0.05,OR=1.975),胃腺癌组含等位基因A的Aa+AA基因型频率也明显高于对照组(P<0.05,OR=2.144);(2)两组ESR2第5外显子RsaⅠ酶切多态基因型rr、Rr、RR频率和等位基因r、R频率的比较,差异均无统计学意义(P>0.05)。结论ESR2第8外显子AluⅠ多态可能与盐城地区青年胃腺癌易感性有关,含等位基因A的Aa和AA基因型可能增加青年胃腺癌发病风险;而ESR2第5外显子RsaⅠ多态与盐城地区青年胃腺癌发病风险无关联。
Objective To investigate the association of estrogen receptor 2 (ESR2) polymorphism with gastric adenocarcinoma in Yancheng area. Methods 100 cases of young patients with gastric adenocarcinoma (gastric adenocarcinoma) and 100 young patients with gastric ulcer (control group) were enrolled in this study. The exon 5 of ESR2 was digested with restriction endonuclease Rsa Ⅰ by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) Polymorphism and exon 8 AluI digestion polymorphism were analyzed. Results (1) AluI polymorphism analysis of exon 8 of ESR2 showed that the frequency of allele A in gastric adenocarcinoma was significantly higher than that in control (P <0.05, OR = 1.975) (2) The frequency and allele of rr, Rr, RR of Rsa Ⅰ digested polymorphism of exon 5 of ESR2 in both groups were significantly higher than those in control group (P <0.05, OR = 2.144) r, R frequency comparison, the difference was not statistically significant (P> 0.05). Conclusion The Alu Ⅰ polymorphism of exon 8 of ESR2 may be associated with the susceptibility to gastric adenocarcinoma in Yancheng area. The genotypes of Aa and AA with allele A may increase the risk of developing gastric adenocarcinoma. However, the Rsa Ⅰ polymorphism of exon 5 of ESR2 No association with the risk of gastric adenocarcinoma in Yancheng area.
