目的:观察诱导分化剂苯乙酸对胶质瘤细胞C6的诱导分化作用,并在分子水平上探讨其作用机理。方法:细胞计数和〔3H〕-TdR掺入法检测细胞增殖,流式细胞术分析细胞周期。结果:苯乙酸对细胞增殖存在剂量(25mmol/L,50mmol/L)依赖性抑制;免疫细胞化学检测CPM值降低,肿瘤细胞DNA合成减少;细胞周期分析G0/G1期所占比例下降,S期相对升高。结论:苯乙酸可阻抑细胞增殖周期,使细胞周期增殖循环中止于S期中DNA合成准备期S0期,DNA合成减少,抑制细胞增殖。其作用机理为抑制细胞周期中G1期的RNA和蛋白质合成
OBJECTIVE: To observe the induction and differentiation of glioma cell C6 induced by the differentiation agent phenylacetic acid, and to explore its mechanism of action at the molecular level. METHODS: Cell proliferation was measured by cell counting and [3H]-TdR incorporation, and cell cycle analysis was performed by flow cytometry. RESULTS: Phenylacetate inhibited the proliferation of cells in a dose-dependent manner (25mmol/L, 50mmol/L); immunocytochemistry showed that the CPM value was decreased and the DNA synthesis of tumor cells was reduced; the percentage of G0/G1 phase in cell cycle analysis Decline, S phase relative increase. Conclusion: Phenylacetic acid can inhibit the cell proliferation cycle and stop the cell cycle proliferation cycle in the S0 phase of DNA synthesis preparation phase in S phase, reduce DNA synthesis, and inhibit cell proliferation. The mechanism of action is inhibition of RNA and protein synthesis in the G1 phase of the cell cycle
