目的探讨ERK1/2抑制剂PD98059对人子宫颈癌细胞株Siha细胞增殖和凋亡的影响。方法不同浓度(10μmol/L25μmol/L,50μmol/L,100μmol/L)PD98059作用于Siha,取不同时间点(6h,12h,24h,48h),然后采用MTT法检测细胞的增殖情况,流式细胞仪检测细胞的凋亡情况。结果 PD98059能够抑制Siha细胞增殖,诱导细胞凋亡,并且呈时间-剂量依赖性(P<0.05)。结论 PD98059能够抑制子宫颈癌细胞株Siha细胞增殖,诱导凋亡,可能与PD98059抑制了RAS-RAF-MEK-ERK信号转导途径有关。
Objective To investigate the effect of ERK1 / 2 inhibitor PD98059 on proliferation and apoptosis of human cervical carcinoma Siha cells. Methods PD98059 cells were treated with PD98059 at different concentration (10μmol / L, 25μmol / L, 50μmol / L, 100μmol / L) for different time points (6h, 12h, 24h, 48h). Cell proliferation was detected by MTT assay. Flow cytometry Instrument detection of cell apoptosis. Results PD98059 could inhibit Siha cell proliferation and induce apoptosis in a time-and dose-dependent manner (P <0.05). Conclusion PD98059 can inhibit the proliferation and induce apoptosis of Siha cells, which may be related to the inhibition of RAS-RAF-MEK-ERK signal transduction pathway by PD98059.
