目的:探讨 HSP70单抗及反义寡核苷酸对前列腺癌细胞株 LNCaP和 PC 3m凋亡的诱导作用及差异。方法:用流式细胞术分析了 HSP70单抗( 2μ l)、反义寡核苷酸( 10μ mol/L)及单抗联合反义寡核苷酸分别处理 LNCaP和 PC 3m 24、 48、 72、 96 h后,细胞凋亡的发生率。结果:在 48、 72、 96 h时单抗联合反义寡核苷酸诱导的凋亡发生率高于单抗或反义寡核苷酸单独诱导时的凋亡率,差异显著 (分别为 P0.05);反义寡核苷酸与单抗在 48、 72、 96 h诱导的凋亡率间差异显著 (P0.05)。结论: HSP70单抗与 HSP70反义寡核苷酸联合可以显著诱导前列腺癌细胞 LNCaP和 PC 3m发生凋亡,为生物疗法治疗前列腺癌提出了新途径。
OBJECTIVE: To investigate the induction of HSP70 monoclonal antibody and antisense oligonucleotide on the apoptosis of prostate cancer cell lines LNCaP and PC 3m. Methods: Flow cytometry was used to analyze the effect of LNCaP and PC 3m 24, 48, 72 treated with monoclonal antibody (2μL), antisense oligonucleotide (10μ mol / L) and monoclonal antibody antisense oligodeoxynucleotides , After 96 h, the incidence of apoptosis. Results: The apoptosis rate induced by MAb antisense oligodeoxynucleotides was higher at 48, 72 and 96 h than that induced by MAb or antisense oligodeoxynucleotides alone (P0 .05). The apoptosis rates induced by antisense oligonucleotide and McAb at 48, 72 and 96 h were significantly different (P <0.05). Conclusion: The combination of HSP70 monoclonal antibody and HSP70 antisense oligodeoxynucleotides can significantly induce the apoptosis of prostate cancer cells LNCaP and PC 3m, and provide a new way for biotherapy for prostate cancer.
