目的:观察干扰素诱导蛋白16(IFI 16)在非小细胞肺癌(NSCLC)、癌旁肺组织和肺感染组织中的表达,探讨其在NSCLC发生发展中的作用。方法:获取2012年6月至2013年6月于东南大学附属中大医院胸外科行肺部分切除术且术中送快速冰冻切片病理确定病变性质的NSCLC患者(22例,分别取肿瘤组织及癌旁肺组织)及肺部感染患者(6例)的新鲜手术标本,用逆转录-聚合酶链反应(RT-PCR)及免疫组织化学方法(IHC)分别测定肿瘤组织、感染肺组织及癌旁肺组织中IFI 16 mRNA和蛋白的表达情况。应用SPSS 20.0统计学软件,采用秩和检验的统计方法比较IFI 16表达在肿瘤组织、感染肺组织、癌旁肺组织间的差异。结果:与癌旁肺组织比较,肿瘤组织IFI 16 mRNA和IFI 16蛋白表达增加,差异均有统计学意义(P<0.05),而肿瘤组织与感染肺组织、以及癌旁肺组织与感染肺组织比较,IFI 16 mRNA和IFI 16蛋白表达差异均无统计学意义。结论:IFI 16 mRNA及蛋白在NSCLC组织表达上调,推测IFI 16可能参与了NSCLC的发生发展,但具体机制尚有待进一步研究。
OBJECTIVE: To observe the expression of interferon-inducible protein 16 (IFI 16) in non-small cell lung cancer (NSCLC), paracancer lung tissue and lung infected tissue and to explore its role in the development of NSCLC. Methods: Patients with NSCLC who underwent partial lung resection from June 2012 to June 2013 at the Department of Thoracic Surgery, CUHU Hospital of Southeast University and who underwent rapid frozen biopsy during the operation from June 2012 to June 2013 were enrolled in this study. Twenty-two NSCLC patients with tumor and cancer And lung infection (6 cases). The tumor tissues, lung tissues and adjacent tissues were detected by reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry (IHC) Lung tissue IFI 16 mRNA and protein expression. SPSS 20.0 statistical software was used to compare the differences of IFI 16 expression between tumor tissues, lung tissues and adjacent lung tissues by statistical methods of rank sum test. Results: Compared with adjacent lung tissue, the expression of IFI 16 mRNA and IFI 16 in tumor tissue increased (P <0.05), while the expression of IFI 16 mRNA and IFI 16 in tumor tissues and lung tissues, adjacent lung tissues and lung tissues There was no significant difference in the expression of IFI 16 mRNA and IFI 16 between the two groups. CONCLUSION: IFI 16 mRNA and protein are up-regulated in NSCLC tissues. It is speculated that IFI 16 may be involved in the development of NSCLC. However, the specific mechanism remains to be further studied.
