目的探讨Th17/Th22/Treg细胞及其细胞因子在先兆流产发病中的作用。方法采用流式细胞术检测20例先兆流产组患者和20例正常对照组女性外周静脉血Th17/Th22/Treg细胞频数,采用ELISA双抗体夹心法检测血清中IL-17A、IL-22、IL-23和IL-35细胞因子,RT-PCR检测Th17/Th22/Treg细胞的转录因子(ROR-γt/AHR/Foxp3)的mRNA表达水平。结果先兆流产组的Th17和Th22细胞百分比高于正常对照组;Treg细胞百分比低于正常对照组;Th17/Treg比例失衡,上述差异均有统计学意义(P<0.01)。先兆流产组的IL-17A、IL-22和IL-23表达水平高于正常对照组;IL-35表达水平低于正常对照组;IL-17A/IL-35比例失衡,上述差异均有统计学意义(P<0.01)。先兆流产组外周血中Foxp3的mRNA表达明显低于对照组;AHR和ROR-γt的mRNA表达明显高于正常对照组,上述差异均有统计学意义(P<0.01)。结论 Th17/Th22/Treg细胞失衡可能导致先兆流产发生。
Objective To investigate the role of Th17 / Th22 / Treg cells and their cytokines in the pathogenesis of threatened abortion. Methods The frequency of Th17 / Th22 / Treg cells in peripheral venous blood of 20 women with threatened abortion and 20 normal controls were detected by flow cytometry. The levels of IL-17A, IL-22 and IL- 23 and IL-35 cytokines. The mRNA expression levels of transcription factors (ROR-γt / AHR / Foxp3) in Th17 / Th22 / Treg cells were detected by RT- Results The percentage of Th17 and Th22 cells in threatened abortion group was higher than that of the normal control group. The percentage of Treg cells was lower than that of the normal control group. The imbalance of Th17 / Treg ratio was statistically significant (P <0.01). The levels of IL-17A, IL-22 and IL-23 in threatened abortion group were higher than those in normal control group; the expression level of IL-35 was lower than that in normal control group; the imbalance of IL-17A / IL- Significance (P <0.01). The mRNA expression of Foxp3 in the peripheral blood of threatened abortion group was significantly lower than that of the control group. The mRNA expression of AHR and ROR-γt was significantly higher than that of the normal control group (P <0.01). Conclusion Th17 / Th22 / Treg cell imbalance may lead to threatened abortion.
