目的建立超高效液相色谱串联三重四级杆飞行时间质谱法(UPLC-Triple-TOF/MS)快速分析三叶青Tetrastigma hemsleyanum的化学成分。方法采用Agilent Eclipse Plus-C18色谱柱(100 mm×4.6 mm,1.8μm),流动相为乙腈(0.1%甲酸)-水(0.1%甲酸),梯度洗脱,体积流量1 m L/min,柱温30℃。采用电喷雾离子源负离子模式,全扫描模式采集样品数据,根据高分辨数据并结合二级质谱信息快速鉴定三叶青的化学成分。结果推测了三叶青中51种化学成分,主要包括黄酮类、酚酸类、磷脂、糖酯类和苯磺酸类等,并对磷脂、糖酯和苯磺酸类化合物的裂解规律进行总结。结论通过UPLC-Triple-TOF/MS联用技术,为定性分析三叶青的化学成分提供了一种快速、高效的定性分析方法。
OBJECTIVE To establish a rapid HPLC-Triple-quadrupole time-of-flight mass spectrometry (UPLC-Triple-TOF / MS) system for the rapid analysis of the chemical constituents of Tetrastigma hemsleyanum. Methods The mobile phase consisted of acetonitrile (0.1% formic acid) -water (0.1% formic acid) with gradient elution on an Agilent Eclipse Plus-C18 column (100 mm × 4.6 mm, 1.8 μm) Temperature 30 ℃. The electrospray ionization source negative ion mode and full scan mode were used to collect the sample data. The chemical composition of clover leaf was identified rapidly by high resolution data combined with the second-order MS information. Results The chemical constituents in 51 leaves were speculated, including flavonoids, phenols, phospholipids, sugar esters and benzenesulfonates. The cracking rules of phospholipids, sugar esters and benzenesulfonates were summarized. Conclusion The UPLC-Triple-TOF / MS combined technique provides a rapid and efficient qualitative analysis method for the qualitative analysis of the chemical constituents of clover leaf.
