目的本研究旨在探讨糖痹康对高脂饲料和小剂量链脲佐菌素(Streptozotocin,STZ)诱导的糖尿病周围神经病变(Diabetic Penipheraineuropathy,DPN)大鼠脂代谢和神经结构的影响。方法将糖尿病大鼠模型分为正常组、模型组、弥可保组、糖痹康低、中、高剂量组。干预治疗15周后,测定各组大鼠血清总胆固醇(TC)、甘油三酷(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、游离脂肪酸(NEFA);并用电镜观察坐骨神经的超微结构。结果模型组大鼠血清中HDL-C含量较正常组明显降低(P<0.05),LDL-C、TG、NEFA明显升高(P<0.05);弥可保与糖痹康低、中、高剂量组大鼠血清中HDL-C含量较模型组明显升高,TC、LDL-C、NEFA含量明显降低(P<0.05)。电镜检测结果显示:弥可保和糖痹康组大鼠坐骨神经形态、髓鞘及轴索等均有不同程度的修复,糖痹康疗效比弥可保明显,且与剂量成正相关。结论糖痹康能改善DPN大鼠脂代谢水平,修复其坐骨神经髓鞘,对DPN有较好的神经保护作用。
Objective This study was designed to investigate the effects of Tangbi Kang on lipid metabolism and neural structure of Diabetic Penipheraineuropathy (DPN) induced by high-fat diet and low-dose streptozotocin (STZ). Methods The diabetic rat model was divided into normal group, model group, Mi Kebao group, Tangbi Kang low, medium and high dose group. After intervention for 15 weeks, the levels of total cholesterol (TC), TG, LDL-C, HDL-C and free fatty acids NEFA). The ultrastructure of sciatic nerve was observed by electron microscope. Results The levels of HDL-C in the serum of rats in model group were significantly lower than those in normal rats (P <0.05), LDL-C, TG and NEFA were significantly increased (P <0.05) Compared with the model group, the content of HDL-C in the serum of the rats in the dose group was significantly increased, and the contents of TC, LDL-C and NEFA were significantly decreased (P <0.05). The results of electron microscopy showed that the forms of sciatic nerve, the myelin sheaths and the axons of rats in Mi Kebao and Tangbikang groups were all repaired in varying degrees. The therapeutic effect of Tongbukang was more obvious than that of Mi Kebao and positively correlated with the dosage. CONCLUSION: Bu Bi Kang can improve lipid metabolism of DPN rats and repair its myelinated sciatic nerve, which has good neuroprotective effect on DPN.
