目的研究不同浓度的BMP-4对于脐静脉内皮细胞体外成血管能力的影响并对其作用机制做初步探讨。方法 BrdU参入法检测细胞增殖情况,划痕实验检测细胞迁移情况,基质胶法检测细胞体外成血管能力,Western blot检测phospho ERK1/2表达情况。结果 BMP-4在低浓度时具有抑制HUVEC的增殖,迁移和成血管,然而随着BMP-4浓度的升高,这种抑制作用逐渐减弱,并有恢复至正常水平的趋势。结论不同浓度的BMP-4对于HUVEC的增殖,迁移及成血管能力具有不同的影响,BMP-4可能是通过ERK/MAPK信号通路影响HUVEC成血管能力的。
Objective To study the effects of different concentrations of BMP-4 on the ability of vascular endothelial cells in vitro and its mechanism of action. Methods BrdU incorporation assay was used to detect the proliferation of cells. Scratch assay was used to detect cell migration. Matrigel assay was used to detect the in vitro vascular endothelial growth factor (VEGF) expression. Western blot was used to detect phospho ERK1 / 2 expression. Results BMP-4 could inhibit the proliferation, migration and angiogenesis of HUVECs at low concentration. However, with the increase of BMP-4 concentration, the inhibitory effect gradually decreased and returned to the normal level. Conclusion BMP-4 at different concentrations has different effects on the proliferation, migration and angiogenesis of HUVEC. BMP-4 may affect the ability of HUVECs to form blood vessels through the ERK / MAPK signaling pathway.
