目的:研究肺癌细胞A549、SpcA1、PLA801及Calu3抗脱落凋亡的特性,为研究肺癌细胞抗脱落凋亡的机制提供理论依据。方法:应用形态学观察,流式细胞术结合PI及AnnexinVFITC双标记染色法及Westernblot法观察4株肺癌细胞脱落凋亡的特性。结果:脱落培养12h后,4株肺癌细胞有不同形态学变化;流式细胞仪检测发现Calu3悬浮培养24h后,凋亡率与贴壁培养24h的凋亡率有明显差别而A549等3种肺癌细胞系与贴壁培养的对照组相比较,其凋亡率无显著性差异,Westernblot结果也验证了上述结果结论:肺癌细胞Calu3属于脱落凋亡细胞,而A549、SpcA1PLA801属于抗脱落凋亡细胞。
OBJECTIVE: To study the anti-apoptosis properties of A549, SpcA1, PLA801 and Calu3 in lung cancer cells, and to provide a theoretical basis for studying the anti-apoptosis mechanism of lung cancer cells. Methods: Morphological observation, flow cytometry combined with PI and AnnexinVFITC double staining and Western blot were used to observe the characteristics of apoptosis and apoptosis in four lung cancer cells. Results: Four kinds of lung cancer cells had different morphological changes after 12 h of culture. The results of flow cytometry showed that the apoptosis rate of Calu3 was significantly different from that of adherent culture for 24h Compared with the adherent culture group, there was no significant difference in the apoptosis rate between the cell lines and the control group. Westernblot results also verify the above results: Calu3 lung cancer cells fall off apoptosis cells, while A549, SpcA1PLA801 belong to anti-shedding apoptotic cells.
