目的研究叉头蛋白(Fox M1)和下调因子(GDDR)在幽门螺杆菌相关胃癌中的表达。方法收集和选取2012年1月-2015年12月本院胃镜活检和手术后胃组织标本150份,其中包含正常胃黏膜组织30份,幽门螺杆菌相关慢性胃炎、肠化生、异物增生、胃癌组织各30份,利用TriZol提取试剂提取RNA并进行逆转录,采用实时荧光定量PCR和免疫组织化学法检测FoxM1基因及FoxM1和GDDR在各疾病阶段表达的情况。结果实时荧光定量PCR检测正常胃组织,慢性胃炎、肠化生、异物增生和胃癌组织FoxM1的mRNA表达水平分别为15.51±2.65、32.68±3.81、41.32±4.09、59.88±6.21和88.65±7.92,免疫组织化学法检测FoxM1相对表达量分别为7.62±1.72、23.21±3.11、33.67±3.91、51.35±5.01和79.56±7.05。GDDR表达评分分别为:正常组织>6分,由幽门螺杆菌引起的胃癌评分<4分,其中<2分(27份)占90%。在由正常组织受到幽门螺杆菌感染向胃癌转化过程中,GDDR表达呈下降趋势。讨论幽门螺杆菌是消化性溃疡和慢性胃炎的主要致病菌,也是引发胃癌的一个重要危险因素。FoxM1和GDDR在正常胃组织,幽门螺杆菌引发的胃炎以及胃癌中组织表达存在差异,检测FoxM1和GDDR可作为幽门螺杆菌引发胃癌的重要依据。
Objective To investigate the expression of Fox M1 and downregulation factor (GDDR) in Helicobacter pylori-associated gastric cancer. Methods We collected and selected 150 samples of gastroscopy biopsy and postoperative gastric tissue from January 2012 to December 2015 in our hospital, which included 30 samples of normal gastric mucosa, chronic Helicobacter pylori associated gastritis, intestinal metaplasia, foreign body hyperplasia, gastric cancer The tissues were made up of 30 parts each. RNA was extracted and reverse transcribed using TriZol extraction reagent. The expression of FoxM1 and FoxM1 and GDDR in various disease stages were detected by real-time fluorescence quantitative PCR and immunohistochemistry. Results The mRNA expression of FoxM1 in normal gastric tissue, chronic gastritis, intestinal metaplasia, foreign body and gastric cancer tissues were 15.51 ± 2.65, 32.68 ± 3.81, 41.32 ± 4.09, 59.88 ± 6.21 and 88.65 ± 7.92 respectively by real-time fluorescence quantitative PCR. Histochemical analysis of FoxM1 relative expression levels were 7.62 ± 1.72,23.21 ± 3.11,33.67 ± 3.91,51.35 ± 5.01 and 79.56 ± 7.05. GDDR expression score were: normal tissue> 6 points, gastric cancer caused by Helicobacter pylori score <4 points, of which <2 points (27) accounted for 90%. During the transformation from Helicobacter pylori-infected normal tissues to gastric cancer, the expression of GDDR showed a decreasing trend. Discussion Helicobacter pylori is the main pathogen of peptic ulcer and chronic gastritis and is also an important risk factor for gastric cancer. FoxM1 and GDDR in normal gastric tissue, Helicobacter pylori-induced gastritis and gastric cancer tissue expression differences exist, detection of FoxM1 and GDDR can be used as an important basis for Helicobacter pylori-induced gastric cancer.
