目的 研究SCP对人胃癌细胞生长、抑制和损伤效应,探讨其抗癌作用机制。方法 用不同浓度的SCP加入体外培养的人胃癌细胞(MGC80—3)中,观察加药后细胞生长和有丝分裂指数(MI)的变化;用MTT法检测其细胞增殖抑制作用;用吖啶橙荧光染色方法观察细胞DNA和RNA含量的变化,用透射电镜观察细胞的损伤效应。结果 SCP明显抑制MGC80—3细胞生长,IC50值为1mg/ml;MI于加药后12h较对照组明显降低;60%以上细胞DNA和RNA荧光染色较对照组明显减弱,核DNA由黄色荧光变为绿色荧光,胞质红色荧光几乎消失,电镜下可见核染色质凝集边移,细胞器变性。结论 SCP可有效抑制人胃癌细胞的增殖。
Objective To study the effect of SCP on the growth, inhibition and injury of human gastric cancer cells and to explore its anti-cancer mechanism. Methods Human gastric cancer cells (MGC80-3) cultured in vitro were treated with different concentrations of SCP. The cell growth and mitotic index (MI) were observed after dosing. The inhibition of cell proliferation was detected by MTT assay. Acridine orange fluorescence The changes of cell DNA and RNA content were observed by staining method. The damage effect of cells was observed by transmission electron microscope. Results SCP significantly inhibited the growth of MGC80-3 cells with an IC50 value of 1 mg / ml. MI was significantly reduced 12 h after dosing compared with the control group. DNA and RNA staining in 60% of the cells was significantly weaker than that of the control group. For the green fluorescence, the cytoplasmic red fluorescence almost disappeared, under electron microscopy, nuclear chromatin condensation shift, organelle degeneration. Conclusion SCP can effectively inhibit the proliferation of human gastric cancer cells.
