目的探讨体外不同浓度5-氟尿嘧啶对人SKOV3卵巢癌细胞株细胞增殖和Skp2表达的影响。方法不同浓度5-氟尿嘧啶0、10、20、40和80μg/ml作用于人SKOV3卵巢癌细胞株后72h,通过细胞计数试剂盒-8(CCK-8)法检测各组卵巢癌细胞增殖情况,采用免疫细胞化学染色法和Realtime PCR技术检测各组Skp2蛋白和mRNA水平的变化。结果不同浓度的5-氟尿嘧啶作用于人SKOV3卵巢癌细胞株72h后,随着浓度的增加,细胞增殖受限明显,Skp2蛋白和mRNA表达水平明显降低,并与5-氟尿嘧啶的浓度呈负相关(r=-0.816,P<0.05;免疫组化Skp2:r=-0.926,P<0.05;Realtime PCR mRNA:r=-0.899,P<0.05)。结论 5-氟尿嘧啶能够抑制SKOV3卵巢癌细胞株的增殖,并能抑制Skp2蛋白和mRNA的表达。
Objective To investigate the effects of 5-fluorouracil in vitro on cell proliferation and Skp2 expression in human ovarian cancer cell line SKOV3. Methods The proliferation of ovarian cancer cells was detected by cytometry kit-8 (CCK-8) method after treated with different concentrations of 5-fluorouracil at 0, 10, 20, 40 and 80 μg / The changes of Skp2 protein and mRNA levels in each group were detected by immunocytochemical staining and Realtime PCR. Results After treated with different concentrations of 5-Fluorouracil for 72 hours, the proliferation of SKOV3 ovarian cancer cell line was significantly inhibited. The expression of Skp2 protein and mRNA was significantly decreased and negatively correlated with the concentration of 5-fluorouracil r = -0.816, P <0.05; Immunohistochemistry Skp2: r = -0.926, P <0.05; Realtime PCR mRNA: r = -0.899, P <0.05). Conclusion 5-Fluorouracil can inhibit the proliferation of SKOV3 ovarian cancer cell lines and inhibit the expression of Skp2 protein and mRNA.
