[目的]研究叶绿酸铜钠(CHL)体外抑制RKO肠癌细胞系的作用及机制。[方法]不同浓度CHL作用于RKO细胞,倒置显微镜下观察形态学变化;流式细胞仪观察CHL对RKO肠癌细胞凋亡的诱导作用;免疫组化检测增殖细胞核抗原(PCNA)、细胞周期素D1(Cy-clin D1)、环氧化酶-2(COX-2)的表达。[结果]CHL对RKO细胞有增殖抑制作用,24h IC50值为0.523±0.061mg/ml,48h IC50值为0.314±0.058mg/ml。随着CHL浓度的增高,RKO细胞抑制率明显上升;CHL能诱导RKO细胞G1期阻滞、降低S期,从而抑制肿瘤细胞增殖。CHL对RKO细胞凋亡未见明显影响。CHL能减少RKO细胞PCNA和COX-2蛋白表达,并具有一定的浓度依赖性。[结论]CHL能明显抑制RKO大肠癌细胞的生长,并且呈一定的浓度和剂量效应,抑制作用可能与PCNA、COX-2表达下调及细胞G1期阻滞有关。
[Objective] To investigate the effect and mechanism of copper chlorophyllin (CHL) inhibiting RKO colon cancer cell lines in vitro. [Methods] RKO cells were treated with different concentrations of CHL and the morphological changes were observed under an inverted microscope. Flow cytometry was used to observe the effect of CHL on the apoptosis of RKO cells. The expressions of proliferating cell nuclear antigen (PCNA), cyclin D1 (Cy-clin D1), cyclooxygenase-2 (COX-2) expression. [Result] CHL could inhibit the proliferation of RKO cells at 24 h IC50 of 0.523 ± 0.061 mg / ml and 48 h at IC50 of 0.314 ± 0.058 mg / ml. With the increase of CHL concentration, the inhibition rate of RKO cells increased obviously. CHL could induce the arrest of G1 phase of RKO cells, reduce the S phase, and inhibit the proliferation of tumor cells. CHL had no obvious effect on RKO cell apoptosis. CHL can reduce RKO cell PCNA and COX-2 protein expression, and has a concentration-dependent manner. [Conclusion] CHL can significantly inhibit the growth of RKO cells in a concentration and dose-dependent manner. The inhibitory effect may be related to the down-regulation of PCNA and COX-2 and the arrest of cell cycle in G1 phase.
