目的:建立RP-HPLC同时测定天麻药材中天麻素、腺苷、对羟基苯甲醇和对羟基苯甲醛的含量。方法:采用Kromasil C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-0.1%醋酸水溶液为流动相,梯度洗脱,流速1.0 mL.min-1,检测波长270 nm,柱温35℃。结果:天麻素、腺苷、对羟基苯甲醇和对羟基苯甲醛分别在19.1~383(r=0.999 9),0.620~12.4(r=0.999 9),2.45~49.0(r=0.999 9),0.280~5.63 mg.L-1(r=0.999 6)与峰面积线性关系良好;平均加样回收率(n=9)均在96.7%~97.7%,RSD均小于1.6%。结论:本法准确可靠,分离度好,适用于天麻药材中天麻素、腺苷、对羟基苯甲醇和对羟基苯甲醛的含量测定。
Objective: To establish a RP-HPLC simultaneous determination of gastrodin, adenosine, p-hydroxybenzyl alcohol and p-hydroxybenzaldehyde in Gastrodia elata Blume. Methods: The gradient elution was performed on a Kromasil C18 column (4.6 mm × 250 mm, 5 μm) using methanol-0.1% aqueous acetic acid as the mobile phase at a flow rate of 1.0 mL · min-1 with a detection wavelength of 270 nm and a column temperature of 35 ° C. Results: Gastrodin, adenosine, p - hydroxybenzyl alcohol and p - hydroxybenzaldehyde were found in 19.1-383 (r = 0.999 9), 0.620-12.4 (r = 0.999 9), 2.45-49.0 (r = 0.999 9) ~ 5.63 mg.L-1 (r = 0.999 6). The average recovery was 96.7% -97.7% with RSDs less than 1.6%. Conclusion: This method is accurate and reliable, good separation, suitable for gastrodin, adenosine, p-hydroxybenzyl alcohol and p-hydroxybenzaldehyde content determination.
