目的观察黄连降脂合剂对AS大鼠气体信号分子硫化氢(H2S)调控的影响,并探讨黄连降脂合剂抗动脉粥样硬化(AS)的作用机制。方法采用高脂饲料喂养联合大剂量维生素D3腹腔注射复制大鼠动脉粥样硬化模型。随机分为正常组、模型组、黄连降脂合剂组、氟伐他汀组。12周以后酶法检测血脂,HE染色观察腹主动脉病理变化情况,采用去蛋白法检测H2S含量。结果正常组几乎无表达,中药组和氟伐他汀组组与模型组比较明显升高(P<0.01);中药组与氟伐他汀组比较,H2S水平升高明显(P<0.05)。结论黄连降脂合剂可能通过上调H2S的水平,从而防治AS形成。
Objective To observe the effects of Huanglian Jiangzhi Mixture on the regulation of hydrogen sulfide (H2S) in asthmatic rats and explore the mechanism of action of Huanglian Jiangzhi Mixture on anti-atherosclerosis (AS). Methods Atherosclerosis model was induced by intraperitoneal injection of high fat diet combined with high dose of vitamin D3. Randomly divided into normal group, model group, Coptis hypoglycemic mixture group, fluvastatin group. After 12 weeks, serum lipids were detected by enzyme-linked immunosorbent assay. The pathological changes of abdominal aorta were observed by HE staining. The content of H2S was detected by deproteinization. Results Compared with fluvastatin group, the expression of H2S was significantly increased in the traditional Chinese medicine group and the fluvastatin group (P <0.01). Conclusion Huanglian Jiangzhi Mixture may prevent AS formation by up-regulating the level of H2S.
