目的探讨骨髓间充质干细胞(MSCs)对体外培养的神经干细胞(NSCs)向少突胶质细胞分化的影响。方法以密度梯度离心的方法获得人hMSCs,设立3个实验组,分别是hMSCs+NSCs共培养组、hMSCs条件培养基+NSCs组、NSCs自发分化组(对照组)。通过免疫细胞化学染色及RT-PCR等手段检测不同组中NSCs向少突胶质细胞分化的情况,并探讨其可能的机制。结果与自发分化组相比,共培养组及hMSCs条件培养基作用组NSCs向少突胶质细胞分化的百分率分别为(61.3±5.7)%和(58.4±6.2)%,差异均具有显著性(<0.05);且RT-PCR结果显示,共培养组及hMSCs条件培养基作用组少突胶质细胞相关基因Olig1、Olig2的表达增强。结论 hMSCs促进体外培养的NSCs向少突胶质细胞分化,可能与hMSCs分泌可溶性因子相关。
Objective To investigate the effect of bone marrow mesenchymal stem cells (MSCs) on the differentiation of neural stem cells (NSCs) into oligodendrocytes in vitro. Methods Human hMSCs were obtained by density gradient centrifugation. Three experimental groups were established: hMSCs + NSCs co-culture group, hMSCs conditioned medium + NSCs group and NSCs spontaneous differentiation group (control group). The differentiation of NSCs to oligodendrocytes in different groups was detected by immunocytochemical staining and RT-PCR, and the possible mechanism was explored. Results Compared with the spontaneous differentiation group, the percentages of NSCs differentiated into oligodendrocytes in co-culture group and hMSCs conditioned medium were (61.3 ± 5.7)% and (58.4 ± 6.2)%, respectively, with significant difference <0.05). The results of RT-PCR showed that the expressions of Olig1 and Olig2 in oligodendrocyte-related genes were enhanced in co-culture group and hMSCs conditioned medium group. Conclusion hMSCs promote the differentiation of NSCs into oligodendrocytes in vitro, which may be related to the secretion of soluble factors by hMSCs.
